Selank: A Research Compendium

What Selank Is

Selank is a synthetic heptapeptide (Thr-Lys-Pro-Arg-Pro-Gly-Pro) developed at the Institute of Molecular Genetics of the Russian Academy of Sciences as a stabilized analogue of the immunomodulatory fragment of the endogenous peptide tuftsin. The proline-glycine tail was added specifically to slow enzymatic degradation and extend the molecule’s biological half-life relative to native tuftsin. In the research literature Selank is most often characterized as a peptide-based anxiolytic and neuropeptide whose reported activity spans the GABAergic, monoaminergic and neurotrophic systems.[1] Because it is delivered to research models intranasally as well as parenterally, it is frequently studied as a model compound for non-injectable peptide neuropharmacology.[5]

Anxiolytic-Type Activity

Selank has been examined extensively as a peptide anxiolytic, with reviews summarizing its molecular pharmacology and behavioral profile in animal models of anxiety.[1] In one rodent chronic-stress paradigm, Selank was reported to enhance the anxiety-reducing effect of diazepam.[2]

GABAergic Signaling

Investigators have studied how Selank influences expression of genes involved in GABAergic neurotransmission, both in cultured neuronal cells and in comparison with GABA itself.[7] Selank is also discussed in pharmacology reviews of agents that interact with the GABA system.[8]

BDNF & Neurotrophic Effects

Intranasal Selank has been reported to regulate brain-derived neurotrophic factor (BDNF) expression in the rat hippocampus.[5] A separate study associated Selank with protection against ethanol-induced memory impairment via changes in hippocampal and prefrontal BDNF content.[6]

Exploratory Clinical Investigation

Selank has appeared in exploratory clinical research on anxiety disorders, including comparisons of its reported anxiolytic effect and tolerability versus a benzodiazepine reference agent.[3] Additional work has examined optimization of treatment approaches for anxiety disorders in these research settings.[4]

Neuroimaging-oriented research has applied a functional connectomic approach to characterize how Selank (alongside the related peptide Semax) affects brain network activity.[9]
