What is Gonadorelin?
Gonadorelin is a synthetic linear decapeptide with the sequence pGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2, structurally identical to the naturally occurring hypothalamic hormone gonadotropin-releasing hormone (GnRH/LHRH). It is typically prepared by solid-phase peptide synthesis and features an N-terminal pyroglutamate and a C-terminal glycine amide. It is classified as a GnRH agonist and is used in research as a reference ligand for the GnRH receptor.
How Gonadorelin is studied
In preclinical and research models, gonadorelin binds the GnRH type I receptor (GnRH-R), a seven-transmembrane G protein-coupled receptor expressed on anterior-pituitary gonadotroph cells. Receptor activation couples primarily to Gq/G11, stimulating phospholipase C-beta to generate IP3 and diacylglycerol, mobilizing intracellular calcium and activating protein kinase C. Pulsatile receptor stimulation is associated in research models with release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), whereas continuous exposure is associated with receptor downregulation.
