What is 5-Amino-1MQ?
5-Amino-1MQ is a synthetic small molecule built on a methylquinolinium (quinoline-derived) scaffold; it is a chemical entity, not a peptide. It was characterized in the published literature as a selective, cell-permeable inhibitor of nicotinamide N-methyltransferase (NNMT). It is described and supplied as a research-use-only reference/tool compound and has no approved human therapeutic status.

How 5-Amino-1MQ is studied
NNMT catalyzes transfer of a methyl group from S-adenosylmethionine (SAM) to nicotinamide, producing 1-methylnicotinamide (1-MNA) and S-adenosylhomocysteine, a reaction that draws on the nicotinamide/NAD+ salvage pathway and the SAM methyl-donor pool. In published preclinical cell work, 5-Amino-1MQ inhibition of NNMT was associated with reduced intracellular 1-MNA and increased intracellular NAD+ and SAM. These observations are mechanistic findings in laboratory models only and do not constitute evidence of any human effect.

Research areas
- Investigated as a chemical-biology tool compound for probing NNMT enzyme activity in vitro
- Studied in cultured adipocyte models for effects on NAD+/SAM metabolite levels and lipid accumulation during differentiation
- Used in rodent (diet-induced obesity) models as a research probe of NNMT-related energy metabolism
- Examined in the context of NAD+ salvage-pathway and SAM methyl-cycle biology research
- Referenced in medicinal-chemistry literature on quinoline-derived NNMT inhibitor design
These describe laboratory/preclinical research only. No therapeutic, medical, or efficacy claims are made.
